Wow. Great match-up. In reality, they'd probably just hug each other, but for the sake of this brawl we'll say they are mortal enemies and want to rip each other to pieces. Barney doesn't have much to fight with. He's just so soft and fuzzy. His hits would be pretty useless. About the only thing he could do is smother Big Bird. I would think Big Bird would have a slight advantage with those long legs and his huge beak. He could probably deliver some good kicks and peck Barney's stuffing out with his huge beak.
